چکیده

Transgenic cotton (Gossypium hirsutum L.) with Bacillus thuringiensis Berliner δ-endotoxin genes has been widely adopted for suppression of lepidopteran pests. To account for inconsistency of the endotoxin gene expression, we developed a polymerase chain reaction (PCR) for checking the purity of transgenic cotton plants. A total of eight cry1Ac genes were aligned for the PCR primer design. A DNA fragment was amplified from Bt cotton, sequenced, and confirmed to be a portion of the Bt gene. A total of 150 cotton plants representing five cultivars were examined for the presence of the Bt gene. Results demonstrated that all of these cotton plants harbored the Bt endotoxin gene.

Fitness of Bt‐resistant cabbage loopers on Bt cotton plants

Development of resistance to the insecticidal toxins from Bacillus thuringiensis (Bt) in insects is the major threat to the continued success of transgenic Bt crops in agriculture. The fitness of Bt-resistant insects on Bt and non-Bt plants is a key parameter that determines the development of Bt resistance in insect populations. In this study, a comprehensive analysis of the fitness of Bt-resi...

Temporal Expression Profiles and Bioactivity of Single (bollgard) and Dual-toxin (bollgard Ii) Bt Cotton

Laboratory studies were conducted using ELISA and bioassays (Helicoverpa zea) to investigate temporal Cry protein expression in field-grown single (‘Bollgard’) and dual-toxin (‘Bollgard II’) Bt cotton. In single-toxin Bt cotton, Cry1Ac levels changed significantly as the season progressed, exhibiting ca. 3-fold difference in Cry1Ac expression between the highest date (6.2 ppm—12 June) and the l...

Improving Environmental Loading Assessments of Cry Protein from GM Plants Based on Experimentation in Cotton

Transgenic Bt-cotton expresses insecticidal Cry proteins via Cry genes, introduced from the soil bacterium Bacillus thuringiensis (Bt). Data on levels of Cry proteins expressed in transgenic Bt-crops is important for assessing levels of environmental exposure. During investigations into the rhizosphere impacts of Cry proteins from GM cotton we found that Cry1Ac expression throughout the season ...

Resistance of Trichoplusia ni populations selected by Bacillus thuringiensis sprays to cotton plants expressing pyramided Bacillus thuringiensis toxins Cry1Ac and Cry2Ab.

Two populations of Trichoplusia ni that had developed resistance to Bacillus thuringiensis sprays (Bt sprays) in commercial greenhouse vegetable production were tested for resistance to Bt cotton (BollGard II) plants expressing pyramided Cry1Ac and Cry2Ab. The T. ni colonies resistant to Bacillus thuringiensis serovar kurstaki formulations were not only resistant to the Bt toxin Cry1Ac, as prev...

Potential factors impacting season-long expression of Cry1Ac in 13 commercial varieties of Bollgard® cotton

Thirteen commercial varieties of transgenic Cry1Ac Bacillus thuringiensis Berliner (Bt) cotton were examined across two sites in 2000 for potential factors that impact endotoxin expression. In all cases, two varieties (NuCOTN 33B and DP 458B/RR, Delta & Pineland Co., Scott, MS) expressed more Cry1Ac than the other 11 varieties in various plant structures. These two varieties share the same pare...
